چکیده

Recently, mutations of the transforming growth factor-b receptor type I gene have been reported to occur at high frequency in breast cancer metastases, with all mutations being an identical C to A transversion at nucleotide 1160 of the gene (T. Chen et al., Cancer Res., 58: 4805–4810, 1998). This mutation would result in a serine to tyrosine substitution at codon 387 (S387Y) and would reportedly disrupt receptor function. Because this mutation reportedly occurred at high frequency in breast cancer metastases (42%) and much less frequently in primary breast cancer tumors (6%), this would seem to represent a pivotal genetic alteration in breast cancer progression. To further investigate the possible role of this specific genetic alteration in the progression of breast cancer and other forms of adenocarcinoma, we analyzed 20 breast cancer metastases, 15 lung adenocarcinoma metastases, and 13 colorectal cancer metastases for possible mutations at this site. Using both single-strand conformation polymorphism screening and sequencing, we found no mutations of this gene in any of our samples. Our results suggest the S387Y mutation of the transforming growth factor-b receptor type I gene is not common in these types of human cancers.

Tissue Expression of Prohibition-I and Its Relationship With Prognostic Factors in Breast Cancer

Background & objective: Globally, breast cancer is the most common malignancy among females. Prohibition (PHB)-I, a homologous protein, was initially introduced as a suppressor gene for amplification process. Further, the protein has a key role in the cell cycle and is capable of inhibiting DNA transcription in many cell types. Therefore, its possible role in different...

Influence of the Different Primary Cancers and Different Types of Bone Metastasis on the Lesion-based Artificial Neural Network Value Calculated by a Computer-aided Diagnostic System,BONENAVI, on Bone Scintigraphy Images

Objective(s): BONENAVI, a computer-aided diagnostic system, is used in bone scintigraphy. This system provides the artificial neural network (ANN) and bone scan index (BSI) values. ANN is associated with the possibility of bone metastasis, while BSI is related to the amount of bone metastasis. The degree of uptake on bone scintigraphy can be affected by the type of bone metastasis. Therefore, t...

بررسی رابطه پروتوانکوژن HER2 با فاکتورهای پروگنوستیک سرطان پستان

Background: Breast cancer is the most common female malignancy world wide and in Iran as well and is  the second cause of death due to malignancy after lung cancer. Varieties of factors such as estrogen and progesterone receptors, and axillary lymph node involvement may influence the prognosis and therapeutic  approach. However, mutation in HER2 gene may also affect the prognosis. In this study...

ZFX Overexpression in Breast Cancer Positively Correlates with Metastasis

Background: As the third most frequent cause of cancer death, breast cancer is a common disease worldwide. Most of the patients are being diagnosed in the stage that conventional treatments are not effective, and invasion and metastases lead to death. Therefore, identification of novel molecular markers to improve early diagnosis, prognosis and treatment of the breast cancer is a necessity. Zin...
